For decades, Alzheimer’s disease has often been described as an inevitable consequence of aging. Research from Harvard Medical School, published in 2025 in the journal Nature, has identified a potential new mechanism involved in the disease: the reduction of natural levels of lithium in the brain.

The study suggests that, during the early stages of cognitive decline, beta-amyloid plaques may bind some of the lithium present in the brain, reducing its availability to nerve cells. In experiments conducted by the researchers, lithium deficiency was associated with inflammation, synapse alterations, and increased accumulation of beta-amyloid and tau protein.

Lithium is an element naturally present in the body in minute amounts and may contribute to several processes beneficial to brain health. This, however, does not mean that its deficiency has been proven to be the sole or primary cause of Alzheimer’s, a complex disease influenced by age, genetics, cardiovascular health, lifestyle, and numerous other factors.

Researchers also tested lithium orotate in mouse models of Alzheimer’s disease. At very low doses, the compound reduced some brain changes, improved synapse function, and partially recovered memory in the animals. The doses used were much lower than those used in psychiatric therapy with other lithium salts.

These results are interesting, but it is important to note that they were obtained primarily in mice. Currently, lithium orotate has not been proven to prevent, treat, or reverse Alzheimer’s disease in humans.

The claim that these doses are completely safe also cannot be extended to humans. Lithium can interfere with kidney and thyroid function, alter electrolytes, and interact with various medications. Furthermore, lithium orotate supplements should not be taken alone for the purpose of protecting memory or treating dementia.

The research, however, opens up an important avenue: controlled clinical trials will be needed to determine whether lithium levels can become a useful biomarker and whether low-dose formulations can be effective and safe in humans.

The discovery does not yet represent a cure, but it offers a new hypothesis to be explored in research on brain aging and Alzheimer’s disease.
